METHODOLOGY

Three tests, in order, on every batch

Each method confirms a different property — purity, identity, and stability. A batch that fails any one is rejected.

Method 01

HPLC

High-Performance Liquid Chromatography

A reverse-phase column separates the compound from impurities. A UV detector reads the eluent at the absorption wavelength of peptide bonds (typically 214 nm). Each peak is integrated, area is normalized, and purity is reported as the percentage of total chromatographic area.

  • Resolution: ≤0.1% impurity detection
  • Column: C18 reverse-phase, 4.6 × 250 mm
  • Detector: UV at 214 nm + 280 nm
  • Run time: 30–60 min per sample
Method 02

Mass Spec

High-Resolution Mass Spectrometry

The compound is ionized via electrospray (ESI) and the resulting ions are mass-separated by a quadrupole-orbitrap. Observed mass is compared to theoretical exact mass — a match within 5 ppm confirms molecular identity. Fragmentation patterns rule out close-mass impurities.

  • Resolution: 70,000 FWHM at m/z 200

Method 03

Stability

Lyophilization & Cold-Chain Verification

Each batch is freeze-dried under controlled vacuum to a residual moisture target below 2%. Karl Fischer titration confirms moisture content. Vials are sealed under nitrogen, then a sample subset is held at -20°C, 4°C, and 25°C for periodic re-test against the original COA.

    Inbound vials are weight-checked, photographed, and barcoded into our LIMS.

    HPLC Screen
    First-pass purity test. Failures are quarantined for re-test or rejected.

    MS Confirmation
    Independent identity confirmation via high-resolution mass spec.
  5. 05
    Moisture & Sterility
    Karl Fischer + bioburden testing on a representative sample.
  6. 06
    COA Issuance
    Lab generates the Certificate of Analysis. QC lead signs off.
  7. 07
    Cold Storage
    Approved batch moves to -4°F storage until packed for shipment.

Volumetric Karl Fischer titrator for residual moisture analysis. Resolution to 1 ppm; certified annually.

SP Scientific VirTis BenchTop Pro

Lyophilizer with -85°C condenser and shelf temperature control. Residual moisture profiles logged for every cycle.
